Biological Background: CD5 Function and Localization
- CD5, also known as Lymphocyte antigen 1 (Ly-1/Lyt-1), is a lymphoid-specific transmembrane glycoprotein receptor expressed on all T-cells and the B1a subset of B-cells.
- It plays a critical role in regulating TCR and BCR signaling, thymocyte selection, T-cell effector differentiation, and immune tolerance.
Related references:
PMID:35720357 - CD5 mediates T-dependent B-cell activation by interacting with B-cell ligands such as CD5L or CD72, and contributes to regulatory T and B-cell homeostasis.
Related references:
PMID:24950378, PMID:9064341, PMID:9723705 - It functions as a negative regulator of TCR signaling during thymocyte development by recruiting signaling proteins including LCK, CD3Z chain, PI3K, and CBL.
Related references:
PMID:11157848 - CD5 is a single-pass type I membrane protein localized at the cell surface.
- The protein features a signal peptide, an extracellular region with disulfide bonds and N-linked glycosylation sites, and a cytoplasmic tail subject to phosphorylation, consistent with its observed molecular weight of ~54 kDa.
Experimental Guidance and Technical Tips
- This APC-conjugated monoclonal antibody is designed for flow cytometry, targeting the extracellular domain (aa 25-371) of mouse CD5. It is suitable for surface staining of live cells without permeabilization.
- For optimal resolution, titrate the antibody concentration using a known positive control (e.g., mouse splenocytes or thymocytes) to determine the best staining index for your experimental setup.
- Since CD5 is expressed on all T-cells and B1a B-cells, include appropriate lineage markers (e.g., CD3, CD19) to distinguish T-cell and B-cell subsets in multi-color panels.
- Avoid using other APC-conjugated antibodies in the same panel to prevent spectral overlap; consider using PE or FITC conjugates for co-staining.
- Validate staining specificity by including an isotype control or by using CD5-knockout cells if available.
