Biological Background: CD4 Function and Localization
- CD4 is a T-cell surface glycoprotein belonging to the immunoglobulin superfamily, also designated as L3T4 or Ly-4 in mice. It functions primarily as a coreceptor during T-cell activation.
- During antigen presentation, CD4 interacts with the T-cell receptor (TCR) and binds to the MHC class II molecule on antigen-presenting cells. This interaction recruits the Src kinase LCK, which phosphorylates downstream signaling proteins, leading to T-helper cell activation, lymphokine production, and adhesion.
- CD4 is essential for the development of T-helper cells in the thymus and also promotes the differentiation of monocytes into mature macrophages.
- The expression of CD4 is a hallmark of T-helper cells but is also detected in other immune cells such as macrophages, dendritic cells, and natural killer cells.
- CD4 is an integral membrane protein with its functional domains exposed on the cell surface, consistent with its role in cell–cell interactions and signaling.
- CD4 is a glycoprotein that undergoes palmitoylation and phosphorylation. It contains disulfide bonds and immunoglobulin-like domains, which are critical for structural integrity and interaction with MHC class II molecules. Alternative splicing generates additional isoforms.
