- CD4 is a T-cell surface glycoprotein also known as L3T4 or Ly-4, encoded by the Cd4 gene in mouse.
- It functions as a coreceptor for MHC class II molecules, interacting with the T-cell receptor (TCR) and MHC II to enhance T-cell activation.
- Upon ligand binding, CD4 recruits the Src kinase LCK to the TCR-CD3 complex, initiating intracellular signaling pathways leading to T-helper cell proliferation, adhesion, and cytokine production.
- In macrophages and NK cells, CD4 mediates differentiation, activation, and migration via TCR-independent mechanisms.
