Biological Background: CD4 Function and Localization
- Encoding the T-cell surface glycoprotein CD4, the Cd4 gene produces a single-pass type I membrane protein with four extracellular immunoglobulin-like domains, a transmembrane helix, and a short cytoplasmic tail. The mouse protein, also known as L3T4 or Ly-4, shares this architecture and plays a central role in adaptive immunity.
- CD4 acts as a coreceptor for MHC class II molecules. On T-helper cells, it binds to the same MHC-peptide complex recognized by the T-cell receptor (TCR), thereby increasing the overall affinity of the interaction and facilitating T-cell activation.
- Upon MHC binding, CD4 recruits the Src-family kinase LCK to the TCR-CD3 complex. This triggers phosphorylation cascades that lead to lymphokine production, T-cell motility, adhesion, and full activation of T-helper cells.
- CD4 is essential for thymic T-helper cell development and for the differentiation of monocytes into functional macrophages. It also participates in activation and cytokine expression in macrophages, dendritic cells, and NK cells, often in a TCR/LCK-independent manner.
- Subcellularly, CD4 is predominantly located at the cell membrane, where it associates with lipid rafts and undergoes post-translational modifications including palmitoylation, N-linked glycosylation, and phosphorylation. These modifications regulate its membrane localization and signaling functions.
- Tissue specificity in mouse: highly expressed on T-helper cells, and also present at lower levels on macrophages, dendritic cells, and NK cells. Its expression is a hallmark of the T-helper lineage.
- The protein is a member of the immunoglobulin superfamily and contains multiple disulfide bonds, features that contribute to its structural stability and ability to interact with its ligands.
Experimental Guidance and Technical Tips
- Because the immunogen is a recombinant protein, the antibody may recognize a linear epitope; consider this when performing antigen retrieval or denaturing western blots, though this antibody is recommended for non-denaturing flow cytometry.
- For flow cytometry, titrate the antibody to determine the optimal concentration for your cell population, and include appropriate isotype controls and single-color compensation controls when using APC/Cyanine7 fluorescence.
- Prepare single-cell suspensions from lymphoid organs (spleen, lymph nodes, thymus) or peripheral blood, and block Fc receptors to minimize non-specific binding before staining.
- Validate expression in the relevant mouse strain and tissue; note that CD4 is highly expressed on T-helper cells but may also be detected on subsets of macrophages, dendritic cells, and NK cells.
