Biological Background: CD317/BST2 Function and Localization
- Also known as bone marrow stromal antigen 2, HM1.24 antigen, GREG, Bst-2, CD317, and DAMP-1, encoded by the Bst2 gene in mouse.
- Acts as an interferon-induced antiviral restriction factor that directly tethers nascent enveloped virions to the cell membrane, preventing their release and spread as cell-free particles.
- Targets a broad range of enveloped viruses, including HIV-1, MMTV, MLV, Ebola virus, Lassa virus, and VSV, by physically linking virions to the cell surface.
- Inhibits cell surface proteolytic activity of MMP14, leading to decreased MMP15 activation and subsequent reduction in cell growth and migration.
- Stimulates signaling by LILRA4/ILT7 on plasmacytoid dendritic cells, providing negative feedback to type I interferon production during viral infection.
- Localizes to the Golgi apparatus, trans-Golgi network, plasma membrane, late endosomes, membrane rafts, and the apical surface of polarized epithelial cells.
- In naive mice, expression is specific to type I interferon-producing cells at the protein level, highlighting its role in innate immunity.
