Biological Background: CD317/BST2 Function and Localization

  • Bone marrow stromal antigen 2 (BST2), also known as CD317 or HM1.24 antigen, is a lipid raft-associated membrane protein encoded by the Bst2 gene in mice.
  • Functions as an interferon-induced antiviral restriction factor that blocks the release of diverse enveloped viruses by directly tethering nascent virions to the cell membrane (e.g., HIV-1, MLV, EBOV, VSV).
  • Exhibits a broad antiviral spectrum, restricting retroviruses, filoviruses, arenaviruses, and rhabdoviruses.
  • Localizes to the trans-Golgi network, cell membrane, late endosomes, membrane rafts, and apical cell membrane; also found in the cytoplasm.
  • Specifically expressed on type I interferon-producing cells (plasmacytoid dendritic cells) in naive mice at the protein level.
  • Contains a signal-anchor transmembrane domain, coiled-coil region, and a GPI-anchor; post-translational modifications include disulfide bonds, N-glycosylation, and lipidation.
  • Plays a role in organizing the subapical actin cytoskeleton in polarized epithelial cells and can inhibit MMP14-mediated cell growth and migration.
  • Provides negative feedback on IFN production by stimulating LILRA4/ILT7 signaling in plasmacytoid dendritic cells.

Experimental Guidance and Technical Tips

  • The immunogen used (amino acids 55-152 of mouse BST2) corresponds to the extracellular domain, which may be suitable for detecting surface expression by flow cytometry.
  • Since this antibody is directly conjugated to , no secondary antibody staining is needed; consider using appropriate isotype control conjugates to ensure specificity.
  • Validate staining in known BST2-expressing mouse cell types, such as plasmacytoid dendritic cells or type I interferon-producing cells.
  • For flow cytometry, titrate the antibody to determine optimal concentration for your specific sample and avoid non-specific binding.
