Biological Background: CD274/PD-L1 Function and Localization

  • CD274, also known as PD-L1 or B7-H1, is a type I transmembrane protein belonging to the immunoglobulin superfamily.
  • Acts as a ligand for the inhibitory receptor PD-1 (PDCD1) to play a critical role in maintaining immune tolerance by modulating T-cell activation thresholds and limiting effector responses. Related references: PMID:11238124
  • Via a yet unknown activating receptor, may costimulate T-cell subsets that predominantly produce interleukin-10 (IL-10). Related references: PMID:11015443 PMID:12719480
  • The PD-1/PD-L1 pathway is exploited by tumors to attenuate anti-tumor immunity; interaction with PD-1 inhibits cytotoxic T lymphocyte (CTL) effector function. Related references: PMID:12218188
  • Blockade of PD-1/PD-L1 signaling reverses T-cell exhaustion and normalizes anti-tumor responses, providing the basis for immune checkpoint cancer immunotherapy. Related references: PMID:12218188
  • Localizes primarily to the cell membrane, and can recycle via early and recycling endosomes.
  • Highly expressed in heart, thymus, skeletal muscle, and lung; weakly in kidney, spleen, thyroid, liver. Expressed on activated dendritic cells, B-cells, macrophages, and numerous tumor cell lines of lymphoid origin.

Experimental Guidance and Technical Tips

  • For Western blot, the predicted molecular weight is ~33 kDa, but glycosylation may result in a band around 40–50 kDa. Validate in the relevant sample system.
  • In IHC-P, consider antigen retrieval and signal amplification due to variable expression levels; validate with appropriate positive and negative tissues.
  • For flow cytometry, use on viable cells to detect surface PD-L1; include appropriate isotype controls and confirm detection in your cellular system.
  • This antibody recognizes mouse CD274; cross-reactivity with other species has not been tested. Confirm if required for your experimental model.
