Biological Background: CD25 Function and Localization
- Interleukin-2 receptor subunit alpha (CD25), also known as p55 or Ly-43, is the high-affinity IL-2 receptor alpha chain.
- CD25 forms a heterotrimeric receptor complex with IL-2 receptor beta (CD122) and common gamma chain (CD132) to bind interleukin-2 with high affinity.
- Essential for regulatory T cell (Treg) function; involved in maintaining immune tolerance by controlling Treg activity and suppressing autoreactive T-cell expansion.
- Localized to the plasma membrane as a type I transmembrane protein, with an extracellular region containing Sushi domains and a signal peptide.
- Post-translational modifications include N-linked glycosylation and disulfide bond formation.
- Belongs to the type I cytokine receptor family; characterized by a transmembrane helix and intracellular signaling motifs.
Experimental Guidance and Technical Tips
- The antibody is generated against a recombinant fusion protein encompassing amino acids 22–236 of mouse CD25, which corresponds to the full extracellular domain. This target region includes the IL-2 binding site and Sushi domains.
- For flow cytometry, it is recommended to titrate the antibody to determine the optimal staining concentration for each cell type and experimental condition.
- As the antibody is conjugated to , no secondary antibody is required; ensure the flow cytometer has appropriate excitation and emission filters (e.g., 561 nm laser, 610/20 nm bandpass).
- Use appropriate isotype controls and include Fc receptor blocking steps to minimize nonspecific binding, especially when analyzing primary mouse immune cells.
- The antibody is validated for mouse reactivity; cross-reactivity with other species has not been tested.
