Biological Background: Mouse CD23 Function and Localization
- Mouse CD23, also known as Fc-epsilon-RII or lymphocyte IgE receptor, is a low-affinity receptor for immunoglobulin E (IgE) and complement receptor 2 (CR2/CD21).
- It plays a pivotal role in regulating IgE production and B-cell differentiation, and facilitates IgE-dependent antigen uptake and presentation to T cells.
- On macrophages, IgE-bound CD23 cross-linking activates the L-arginine–nitric oxide pathway, leading to intracellular killing of parasites.
- The receptor exists as both membrane-bound and secreted isoforms via alternative splicing, localizing primarily to the cell membrane but also found in a soluble form.
- CD23 is a type II transmembrane glycoprotein modified by palmitoylation, disulfide bonds, and glycosylation; it features a C-type lectin domain, calcium-binding sites, and a proteoglycan form.

- The immunogen spans the extracellular domain (aa 50–331), so the antibody is expected to recognize both membrane-bound and soluble CD23.
