Biological Background: Lymphocyte activation gene 3 protein Function and Localization
- LAG-3 functions as an inhibitory receptor on antigen-activated T-cells, delivering suppressive signals upon engagement with its ligands MHC class II on antigen-presenting cells and FGL1 secreted by hepatocytes and tumor cells. Related references: PMID:12209638, PMID:12421911, PMID:12672063, PMID:30349037
- Ligand binding induces ubiquitination of the KIEELE motif, releasing the RRFSALE motif and promoting the formation of condensates with the TCR component CD3E, thereby disrupting CD3E-LCK association and inhibiting TCR signaling. Related references: PMID:40101708, PMID:40592325
- LAG-3 may synergize with PD-1/PDCD1 to inhibit antigen-specific T-cell activation. Related references: PMID:21300912
- It negatively regulates proliferation, activation, effector functions, and homeostasis of both CD8+ and CD4+ T-cells. Related references: PMID:12209638, PMID:12421911, PMID:12672063
- Constitutively expressed on a subset of regulatory T-cells (Tregs), LAG-3 contributes to their suppressive function and also acts as a negative regulator of plasmacytoid dendritic cell (pDC) activation. Related references: PMID:15485628, PMID:19201841
- The LAG-3 inhibitory pathway is exploited by tumors to attenuate anti-tumor immunity, highlighting its role as an immune checkpoint target.
- Primarily expressed in activated CD4+ and CD8+ T-cells, with additional expression in a subset of Tregs and pDCs. Related references: PMID:12209638, PMID:15485628, PMID:19201841
- LAG-3 localizes to the cell membrane and can also be secreted.
Experimental Guidance and Technical Tips
- When using this -conjugated antibody for flow cytometry, consider titrating on activated mouse splenocytes or T-cell lines to determine optimal staining concentrations.
- The immunogen covers the extracellular domain (aa 24–442), which may ensure recognition of the native receptor; verify performance on live, unfixed cells.
- As LAG-3 is an activation marker, in vitro stimulation of T-cells (e.g., with anti-CD3/CD28) may be required to upregulate expression before analysis.
- For intracellular staining, evaluate compatibility with fixation/permeabilization protocols, as the fluorophore may be sensitive to certain buffers.
- Include appropriate isotype controls and, if multiplexing, check for potential compensation issues with the dye.
