Biological Background: CD22 Function and Localization
- CD22 (Siglec-2, BL-CAM) is the most highly expressed siglec on B-cells, playing a critical role in B-cell differentiation, antigen presentation, and bone marrow trafficking. Related references: PMID:7533044 PMID:9016707
- It functions as an inhibitory coreceptor of the B-cell receptor (BCR), dampening BCR-induced calcium mobilization and cellular activation. Related references: PMID:9016707
- The immunoreceptor tyrosine-based inhibitory motif (ITIM) of CD22 is phosphorylated by Src family kinase LYN, leading to recruitment of protein tyrosine phosphatase SHP-1 (PTPN6) and negative regulation of BCR signaling.
- Strong negative signaling through CD22 can induce B-cell apoptosis.
- CD22 binds α2,6-linked sialic acid residues on cis surface molecules (CD22, CD45, IgM) and may mediate trans cell adhesion through sialic acid binding.
- The protein is expressed specifically in B-lymphocytes and localizes to the cell membrane.
- Alternative splicing generates multiple isoforms; the canonical isoform of mouse CD22 has a predicted molecular weight of approximately 97.2 kDa (UniProt P35329).
Experimental Guidance and Technical Tips
- The immunogen (aa 22–151) targets the extracellular N-terminal region, including the Ig-like V-set domain, suggesting the antibody can recognize native surface CD22 on mouse B-cells in flow cytometry.
- For best results, titrate the antibody and include appropriate isotype controls and Fc receptor blocking steps when staining primary B-cells or B-cell lines.
- CD22 expression is restricted to B-lymphocytes; validate specificity using B-cell positive and T-cell/other lineage negative controls.
- The conjugate is compatible with common violet/UV laser lines; avoid buffers containing high autofluorescence or preservatives that may interfere with the dye.
- Ship and store under ice bag conditions to maintain conjugate stability; protect from light.
