Biological Background: CD150/SLAM Function and Localization
- CD150/SLAM (SLAMF1) is a self-ligand receptor of the signaling lymphocytic activation molecule family, which modulates the activation and differentiation of a wide variety of immune cells, integrating both innate and adaptive immunity.
- In T-cells, SLAMF1 mediates IL-2-independent proliferation, induces IFN-gamma production, and promotes TCR-induced IL-4 secretion by CD4+ cells; it is essential for IL-4 production by germinal center T follicular helper cells. Related references: PMID:12351401 PMID:9126961 PMID:15123745 PMID:20525889
- Its downstream signaling involves both SH2D1A/SAP-dependent and independent pathways, with divergent outcomes: the INPP5D-DOK1-DOK2 axis inhibits IFN-gamma, while PRKCQ-BCL10-NFKB1 promotes T-cell activation and Th2 cytokine production. Related references: PMID:11477403 PMID:15539155 PMID:16847311
- In conjunction with SLAMF6, controls the transition from positive selection to expansion and differentiation of thymic NKT cells, and drives peripheral iNKT cell differentiation toward a regulatory NKT2 phenotype. Related references: PMID:18031695 PMID:18606638
- In B-cells, SLAMF1 activates ERK and Akt signaling independently of SAP but dependent on SYK and INPP5D; together with CD84/SLAMF5 and SLAMF6, it may serve as a negative regulator of humoral immune responses. Related references: PMID:15315965 PMID:25926831
- In macrophages, SLAMF1 down-regulates IL-12, TNF, and nitric oxide in response to LPS, and mediates innate immune defense against Gram-negative bacteria by recognizing bacterial surface proteins (likely OmpC/OmpF), regulating phagosome maturation and PI3K complex II recruitment. Related references: PMID:15123745 PMID:20818396 PMID:22493499
- May also regulate allergen-induced Th2 and Th1 cytokine secretion, implicating a role in allergic diseases. Related references: PMID:16528012
- A transmembrane glycoprotein expressed on the cell membrane, consistent with its role in cell adhesion and signaling; it contains immunoglobulin-like domains and undergoes alternative splicing and phosphorylation.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the extracellular domain (aa 25-242) of mouse CD150/SLAM, indicating the antibody likely an extracellular epitope, making it suitable for detecting surface expression in live cells by flow cytometry.
- As a directly conjugated monoclonal antibody, it is optimized for flow cytometry; consider titrating to determine the optimal staining concentration for your specific cell population.
- Validate expression in relevant primary cells such as activated T cells, B cells, dendritic cells, macrophages, or NKT cells, where CD150/SLAM is known to be expressed.
- Include appropriate isotype controls and live/dead discrimination to ensure specific staining.
- Protect the antibody conjugate from light and avoid repeated freeze-thaw cycles to maintain fluorophore stability.
