Biological Background: CD130/gp130 (IL6ST) Function and Localization

  • CD130/gp130, encoded by the Il6st gene, is the interleukin-6 receptor subunit beta, also known as IL-6 signal transducer or membrane glycoprotein gp130 (UniProt entry Q00560).
  • It functions as a shared signal-transducing receptor for multiple cytokines including IL-6, LIF, OSM, CNTF, IL-11, CT-1, and CLCF1. Upon cytokine binding, gp130 homodimerizes or heterodimerizes with LIFR or OSMR, activating JAK/STAT signaling pathways.
    Related references:
    PMID:1602143
    PMID:10661409
  • The IL-6 signaling pathway induces expression of SOCS1 and SOCS3, which negatively regulate JAK activity as a feedback mechanism.
    Related references:
    PMID:9202125
  • Gp130 also activates YAP and NOTCH pathways to control inflammation-induced epithelial regeneration, independent of STAT3.
    Related references:
    PMID:25731159
  • It plays critical roles in immune response, hematopoiesis, pain modulation, bone metabolism, and embryonic development. It is essential for survival of motor and sensory neurons and astrocyte differentiation.
    Related references:
    PMID:10661409
    PMID:25057188
    PMID:26255596
    PMID:10377352
    PMID:25228504
    PMID:24339143
    PMID:9348227
  • Subcellularly, gp130 is a single-pass type I membrane protein located at the cell membrane.
  • It is widely expressed in various tissues including brain, heart, thymus, spleen, kidney, lung, and liver, and is found in many cell lines.
    Related references:
    PMID:28402851
  • Post-translational modifications include disulfide bonds, glycosylation, and phosphorylation, and it contains immunoglobulin-like domains and fibronectin type III repeats.
  • Required for expression of TRPA1 in nociceptive neurons and maintenance of PTH1R in osteoblasts, influencing pain sensation and bone formation.

    PMID:25057188
    PMID:25228504

Experimental Guidance and Technical Tips

  • This -conjugated monoclonal antibody is designed for direct flow cytometry (FC) applications, eliminating the need for a secondary antibody.
  • The immunogen corresponds to the extracellular domain (aa 23-617) of mouse CD130/gp130, so this antibody should recognize the native cell surface protein.
  • Since gp130 is widely expressed, include appropriate positive and negative controls (e.g., knockout cells or isotype controls) to confirm specificity.
  • For optimal staining, titrate the antibody in the relevant sample system and consider a viability dye to exclude dead cells.
  • has excitation/emission maxima around 590/617 nm; verify compatibility with your flow cytometer's laser and filter configuration.
