Biological Background: Integrin alpha-M (CD11b) Function and Localization
- Integrin alpha-M (CD11b, encoded by Itgam) is the alpha subunit of the ITGAM/ITGB2 (Mac-1/CR3) integrin complex and belongs to the integrin alpha chain family.
- It serves as a receptor for the iC3b fragment of complement component C3, mediating uptake of complement-coated particles and pathogens.
- The complex also binds fibrinogen, factor X, and ICAM1, facilitating leukocyte adhesion and neutrophil migration.
- In microglia, ITGAM/ITGB2 cooperates with TYROBP/DAP12 to regulate superoxide production, which promotes neuronal apoptosis during brain development. Related references: PMID:18685038
- Predominantly expressed in monocytes, granulocytes, and microglia, with specific expression in a subset of peritoneal mast cells. Related references: PMID:3887182 PMID:8986723 PMID:9862668 PMID:18685038
- Subcellular localization: cell membrane and membrane rafts.
- Features multiple disulfide bonds, glycosylation, and metal-binding (calcium) sites typical of integrins.
Experimental Guidance and Technical Tips
- For flow cytometry, titrate the antibody to determine the optimal working concentration for your specific cell type and experimental conditions.
- The conjugate is photolabile; protect from light during handling and storage.
- Validate the antibody in your sample system using appropriate controls, as cross-reactivity is confirmed only for mouse.
- Upon receipt, store at 2–8°C and protect from light; avoid freeze-thaw cycles.
