Biological Background: CD11a Function and Localization
- Integrin alpha-L (CD11a) is the alpha subunit of the leukocyte-specific integrin LFA-1, which heterodimerizes with the beta-2 integrin subunit (ITGB2/CD18). It is a type I transmembrane glycoprotein containing an extracellular domain with several calcium-binding repeats.
- LFA-1 functions as a receptor for intercellular adhesion molecules (ICAM1, ICAM2, ICAM3, ICAM4) and for F11R, mediating leukocyte-endothelial interactions, T-cell killing, and antibody-dependent cellular cytotoxicity.
Related references: PMID:2051027 - It also binds to the secreted ubiquitin-like protein ISG15, modulating immune responses.
Related references: PMID:29100055 - LFA-1 is essential for leukocyte adhesion, transendothelial migration, and the formation of the immunological synapse, translating TCR engagement into graded adhesion.
Related references: PMID:16234355, PMID:24158516 - Required for lymphopoiesis, specifically the generation of common lymphoid progenitor cells in bone marrow.
Related references: PMID:25108025 - Contributes to natural killer cell cytotoxicity and, in association with ICAM3, facilitates macrophage phagocytosis of apoptotic neutrophils.
- Subcellular localization: Predominantly at the cell membrane and enriched in membrane rafts on leukocytes.
- Post-translational modifications include N-linked glycosylation, phosphorylation, and disulfide bond formation.
Experimental Guidance and Technical Tips
- The immunogen corresponds to an extracellular fragment (amino acids 149–340), suggesting the antibody recognizes an epitope within the extracellular domain. Validate reactivity in relevant mouse leukocyte populations, such as lymphocytes and monocytes, via flow cytometry.
- For FC, determine optimal antibody concentration by titration on positive (e.g., mouse splenocytes) and negative control cells. Include appropriate isotype controls to assess non-specific binding.
- Consider fixation and permeabilization protocols if intracellular staining is required, though CD11a is primarily surface-expressed.
