Biological Background: Lysosome-associated membrane glycoprotein 2 (LAMP-2) Function and Localization

  • LAMP-2, also designated CD107b, Mac3, LGP-B, Lamp II, and Lamp-2 isoforms (Lamp-2a, Lamp-2b, Lamp-2c), is a lysosome-associated membrane glycoprotein encoded by the Lamp2 gene in mice.
  • It plays a critical role in lysosome biogenesis and lysosomal pH regulation by acting as a direct inhibitor of the proton channel TMEM175, thereby facilitating lysosomal acidification for optimal hydrolase activity.
  • LAMP-2 functions in chaperone-mediated autophagy (CMA) by binding target proteins such as GAPDH, GPX4, NLRP3, and MLLT11, and targeting them for lysosomal degradation. Related references: PMID:30718432
  • It is required for the fusion of autophagosomes with lysosomes during autophagy; cells lacking LAMP2 fail to accumulate STX17 on autophagosomes. Related references: PMID:27628032
