Biological Background: MOB4 (MOB-like protein phocein) Function and Localization
- MOB4, also known as phocein, preimplantation protein 3, or MOBKL3, is a member of the MOB family of kinase activator-like proteins.
- It is a core component of the striatin-interacting phosphatase and kinase (STRIPAK) complexes, which regulate protein phosphorylation and dephosphorylation events.
- Through STRIPAK, MOB4 participates in modulating multiple signaling cascades, including the Hippo, MAPK, and nuclear receptor pathways, as well as cytoskeleton remodeling.
- STRIPAK complexes containing MOB4 are implicated in diverse biological processes such as cell growth, differentiation, apoptosis, metabolism, and immune regulation.
- MOB4 localizes predominantly to the cytoplasm and perinuclear region, and is also found associated with the Golgi apparatus and Golgi stack membranes.
- The protein is subject to phosphorylation and binds metal ions such as zinc, features consistent with its regulatory roles.
- Alternative splicing of the MOB4 gene generates multiple transcript variants, indicating potential isoform diversity.
Experimental Guidance and Technical Tips
- For Western blot, the expected band is approximately 26 kDa. Validate with appropriate positive controls, as expression levels may vary.
- In immunohistochemistry (paraffin-embedded sections), optimize antigen retrieval and consider the perinuclear/Golgi staining pattern.
- For ELISA, pair with a matched detection antibody or antigen, and titrate antibody concentration.
- The antibody is raised against a human sequence but cross-reacts with mouse and rat; verify cross-reactivity in your sample system.
- As a polyclonal antibody, lot-to-lot variability may occur; validate each new lot.
