Biological Background: MOB1A/MOB1B Function and Localization

  • MOB1A (Mps one binder kinase activator 1A) and MOB1B are core components of the Hippo signaling pathway, acting as activators of the LATS1/2 kinases.
  • MOB1 forms a regulatory complex with LATS1/2, which is phosphorylated by MST1/2 (STK4/STK3) kinases, leading to LATS activation and subsequent phosphorylation and inactivation of the oncoproteins YAP1 and WWTR1/TAZ.
  • Phosphorylation of YAP1/TAZ by LATS1/2 prevents their nuclear translocation, inhibiting the transcription of genes that promote proliferation and migration, thereby contributing to organ size control and tumor suppression.
  • MOB1A also stimulates the kinase activity of STK38 and STK38L, and cooperates with STK3/MST2 to activate STK38.
  • MOB1A is widely expressed across tissues, including adrenal gland, bone marrow, brain, placenta, prostate, salivary gland, skeletal muscle, testis, thymus, thyroid gland, heart, spinal cord, fetal brain, and fetal liver.
  • Post-translational modifications include acetylation and phosphorylation; MOB1A is a zinc-binding phosphoprotein.
  • The protein undergoes alternative splicing and has been identified through direct protein sequencing and proteomics.

Experimental Guidance and Technical Tips

  • Due to sequence homology, this polyclonal antibody is likely to detect both MOB1A and MOB1B. Validate specificity using knockdown or overexpression models in the relevant species.
  • For Western blotting, consider using a dilution optimized for the target abundance in your sample; a band around 25 kDa is expected.
  • IF/ICC applications may require optimization of fixation and permeabilization conditions; include appropriate positive and negative controls.
  • ELISA performance can be affected by coating buffer and blocking conditions; titrate the antibody against the specific antigen to determine the optimal working concentration.
