Biological Background: MNAT1 Function and Localization
- MNAT1, also known as MAT1, CDK7/cyclin-H assembly factor, Cyclin-G1-interacting protein, Menage a trois, RING finger protein 66 (RNF66), p35, and p36, is a crucial assembly factor for the CDK-activating kinase (CAK) complex.
- It binds and activates CDK7, the catalytic subunit of CAK, which then phosphorylates CDK1, CDK2, CDK4, and CDK6 on a conserved threonine residue, essential for cell cycle progression. Related references: PMID:41100585
- As part of the CAK complex within the TFIIH basal transcription factor, MNAT1 facilitates the phosphorylation of the RNA polymerase II C-terminal domain (CTD), enabling promoter escape and transcription elongation. Related references: PMID:10024882
- MNAT1 localizes to the nucleus, consistent with its roles in cell cycle control and transcription regulation.
- The protein contains a RING finger domain and binds zinc, indicating metal-binding properties; it is also subject to post-translational modifications including acetylation and phosphorylation.
- Tissue expression analysis shows highest MNAT1 levels in colon and testis, with moderate expression in thymus, prostate, ovary, and small intestine, and lowest in spleen and leukocytes.
Experimental Guidance and Technical Tips
- For western blot, expect a band at approximately 36 kDa. Use lysates from colon or testis tissues (high MNAT1 expression) as positive controls, and validate specificity with siRNA knockdown or knockout cell lines.
- Due to the polyclonal nature of this antibody, perform titration experiments to optimize concentrations for IP and ELISA, and consider batch-to-batch variability when switching lots.
