Biological Background: MMP9 Function and Localization

  • Matrix metalloproteinase-9 (MMP9), also known as gelatinase B or 92 kDa type IV collagenase, is a secreted zinc-dependent endopeptidase belonging to the matrix metalloproteinase family. It is synthesized as a zymogen (pro-MMP9) that requires proteolytic activation to become fully active.
  • MMP9 plays a critical role in the degradation of the extracellular matrix (ECM), particularly in the cleavage of type IV and type V collagen into large C-terminal three-quarter fragments and shorter N-terminal one-quarter fragments, as well as fibronectin, but does not degrade laminin or Pz-peptide.
  • Involved in leukocyte migration across basement membranes by facilitating ECM proteolysis, thereby contributing to inflammatory responses and immune cell trafficking.
  • MMP9 participates in bone remodeling by mediating osteoclastic bone resorption, a process essential for normal bone development and pathological bone loss. Related references: PMID:8132709
  • Cleaves NINJ1 (nerve injury-induced protein 1) to generate secreted ninjurin-1, which may play roles in cell adhesion and inflammation. Related references: PMID:23142597, PMID:32883094
  • MMP9 is located in the extracellular space, predominantly associated with the extracellular matrix, where it interacts with other ECM components and inhibitors like TIMPs (tissue inhibitors of metalloproteinases).
  • The enzyme contains characteristic structural domains including a signal peptide, a pro-domain that maintains latency, a catalytic domain with a zinc-binding motif, and a hemopexin-like domain involved in substrate recognition and enzyme localization; it also requires calcium for structural stability.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to amino acids 108-730 of mouse MMP9, a region spanning the catalytic and hemopexin domains, which suggests the antibody may recognize both pro- and active forms. For WB, a band at approximately 80 kDa (pro-form) and possibly lower bands for cleaved active species may be observed. Validate in the relevant sample system.
  • This rabbit polyclonal antibody is suitable for Western blot (WB) and ELISA. For WB, consider using reducing conditions and include appropriate controls. For ELISA, the antibody can be used as capture or detection reagent; optimize coating and detection concentrations for your specific assay.
  • Cross-reactivity is validated for mouse MMP9. When using tissues or cells from other species, check sequence homology and perform a pilot experiment.
  • Store the antibody as recommended (ice bag shipping indicates cold storage) and avoid repeated freeze-thaw cycles to maintain activity.
