Biological Background: MMP3 Function and Localization

  • MMP3 (Stromelysin-1) is a secreted zinc-dependent metalloproteinase with broad substrate specificity, degrading fibronectin, laminin, gelatins, collagens (III, IV, X, IX), and cartilage proteoglycans; it also activates growth factors, plasminogen, and other MMPs such as MMP9.   • The inactive pro-enzyme is released into the extracellular matrix and activated via the plasmin cascade signaling pathway. Related references: PMID:2383557

  • The immunogen corresponds to the C-terminal region (aa 385-477) of human MMP3. For western blotting, a band around 54 kDa (pro-form) may be observed, with possible processed active forms at lower molecular weights; validate the antibody in your sample system and consider using protease inhibitors.
  • For immunohistochemistry (IHC-P), antigen retrieval using heat-induced epitope retrieval may improve staining; optimize conditions for your tissue type.
  • In immunofluorescence/immunocytochemistry (IF/ICC), permeabilization is essential to detect intracellular MMP3; use appropriate controls to confirm specificity.
  • For ELISA applications, pair this antibody with a suitable capture/detection antibody; test serial dilutions to determine optimal working concentrations.
