Biological Background: Stromelysin-1 Function and Localization
- Stromelysin-1 (MMP-3) is a secreted zinc-dependent endopeptidase belonging to the matrix metalloproteinase (MMP) family, also known as transin-1.
- It degrades a broad spectrum of extracellular matrix (ECM) components, including fibronectin, laminin, gelatins (types I, III, IV, V), collagens (III, IV, X, IX), and cartilage proteoglycans; it also activates other molecules such as pro-MMP9 and growth factors. - In immunohistochemistry (IHC-P), antigen retrieval (heat- or enzyme-mediated) is often required; titrate the antibody and include appropriate positive and negative tissue controls.
- For immunofluorescence (IF/ICC), fix and permeabilize cells under optimized conditions; consider secretion inhibitors if targeting the intracellular pool of MMP3.
