- Matrix metalloproteinase-25 (MMP25), also known as leukolysin and membrane-type matrix metalloproteinase 6 (MT-MMP6), is a member of the matrix metalloproteinase family.
- It is a membrane-anchored protease with a glycosylphosphatidylinositol (GPI) anchor, and can also be secreted into the extracellular space and matrix.
- MMP25 may activate progelatinase A, contributing to extracellular matrix remodeling and tissue turnover.
- Expressed predominantly in leukocytes, lung, and spleen, with elevated levels observed in colon carcinoma, astrocytoma, and glioblastoma, suggesting roles in inflammation and cancer progression.
- The protein contains a propeptide domain, a catalytic domain with a zinc-binding site, and hemopexin-like repeats; its activity depends on calcium and zinc.
- Belongs to the membrane-type MMP (MT-MMP) subfamily, specifically designated MT6-MMP, and shares structural features with other GPI-anchored MMPs.
Experimental Guidance and Technical Tips
- The immunogen spans the hemopexin-like domain (residues 321–539); consider that the antibody may detect both full-length and shed/truncated forms of MMP25.
- In Western blot, the predicted molecular weight is approximately 63 kDa, but proteolytic processing could generate lower bands; validate in your sample system with appropriate controls.
- For IHC-P and IF/ICC, optimize antigen retrieval and permeabilization protocols to account for membrane localization and potential secretion of the target.
- When using mouse or rat tissues, verify cross-reactivity experimentally, as sequence homology may influence detection sensitivity.
