- MMP2, also known as 72 kDa type IV collagenase or gelatinase A, is a member of the matrix metalloproteinase (MMP) family. It is encoded by the MMP2 gene, which undergoes alternative splicing.
- It degrades extracellular matrix components such as type IV collagen and gelatin, and also acts on nonmatrix proteins including big endothelin-1 and beta-type CGRP, promoting vasoconstriction.
- MMP2 plays critical roles in tissue remodeling, angiogenesis, tumor invasion, inflammation, and atherosclerotic plaque rupture.
- The C-terminal non-catalytic fragment (PEX) possesses anti-angiogenic and anti-tumor properties, inhibits cell migration, and serves as a ligand for integrin αvβ3 on blood vessels.
- MMP2 mediates proteolysis of CHUK/IKKA, initiating innate immune signaling via NF-kappaB, NFAT, and IRF pathways in response to mitochondrial-nuclear stress.
- Subcellular localization includes secretion into the extracellular space, association with the extracellular matrix and membrane, and presence in the nucleus, cytoplasm, and mitochondria.
