Biological Background: MMP2 Function and Localization

• MMP2 encodes the 72 kDa type IV collagenase, also known as gelatinase A or matrix metalloproteinase-2 (MMP-2), a member of the zinc-dependent metalloproteinase family.
• It functions ubiquitously in extracellular matrix degradation and remodeling, critical for physiological processes like angiogenesis, tissue repair, and vasculature remodeling, as well as pathological conditions including tumor invasion, inflammation, and atherosclerotic plaque rupture.
• Beyond ECM proteins, MMP2 cleaves nonmatrix substrates such as big endothelin-1 and beta-type CGRP, promoting vasoconstriction, and also processes KISS.

• For Western blotting (WB), a band around 74 kDa is expected (pro-form), with lower bands possible for active forms; validate in your sample system.
• For IF/ICC, note that MMP2 is secreted but also found intracellularly; permeabilization may be required, and signal may be detected in the cytoplasm, nucleus, or mitochondria depending on cell type and conditions.
• Cross-reactivity with mouse and rat is reported; for other species, verify sequence homology in the immunogen region.
• ELISA: The antibody can be used for quantitative detection, but appropriate controls and standard curves should be established.
