Biological Background: MMP14/MT1-MMP Function and Localization
- MMP14/MT1-MMP is a membrane-type matrix metalloproteinase belonging to the MMP family.
- Degrades various extracellular matrix components, including collagen, and is essential for pericellular collagenolysis and tissue remodeling. Related references: PMID:8015608
- Activates progelatinase A (pro-MMP2), positively regulating cell growth and migration, and participates in fibrovascular tissue formation. Related references: PMID:8015608, PMID:12714657, PMID:22065321
- Regulates other cellular processes: actin cytoskeleton reorganization via PTK7 cleavage, Notch signaling via DLL1 cleavage, angiogenesis inhibition via ADGRB1 cleavage, and GDF15-GFRAL aversive response via GFRAL cleavage. Related references: PMID:20837484, PMID:21572390, PMID:22330140, PMID:35177851
- Subcellular localization includes the cell membrane, melanosomes, and cytoplasm.
- Expressed in stromal cells of colon, breast, and head and neck, and in lung tumors.
- Key structural features: calcium- and zinc-dependent metalloprotease; synthesized as a zymogen and activated by proteolytic cleavage; subject to phosphorylation.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide from amino acids 75-174 of human MMP14; the antibody may recognize both the pro-form and the active form.
- Validated for IF/ICC and ELISA. For other applications like WB or IHC, consider further validation in your experimental system.
- Based on tissue expression, consider using colon, breast, or head and neck cancer cell lines or tissues as positive controls.
- For IF/ICC, use appropriate fixation and permeabilization methods to detect membrane and cytoplasmic localization.
- MMP14 is often upregulated in invasive cancers; consider its role in cell migration and ECM remodeling in your experimental design.
