Biological Background: MMP14/MT1-MMP Function and Localization
- Membrane-type matrix metalloproteinase, also known as MMP-X1, MT-MMP 1, MT1-MMP, and MT1MMP.
- Endopeptidase that degrades various components of the extracellular matrix such as collagen; essential for pericellular collagenolysis and modeling of skeletal and extraskeletal connective tissues during development. Related references: PMID:8015608
- Activates progelatinase A/MMP2, acting as a positive regulator of cell growth and migration, and is involved in the formation of fibrovascular tissues in association with pro-MMP2. Related references: PMID:22065321, PMID:8015608, PMID:12714657
- May be involved in actin cytoskeleton reorganization by cleaving PTK7. Related references: PMID:20837484
- Acts as a regulator of Notch signaling by mediating cleavage and inhibition of DLL1. Related references: PMID:21572390
- Cleaves ADGRB1 to release vasculostatin-40, which inhibits angiogenesis. Related references: PMID:22330140
- Acts as a negative regulator of the GDF15-GFRAL aversive response by mediating cleavage and inactivation of GFRAL. Related references: PMID:35177851
- Localizes to the cell membrane, melanosomes, and cytoplasm. Expressed in stromal cells of colon, breast, and head and neck; also found in lung tumors.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide covering amino acids 100-200 of human MMP14, which may detect both pro and active forms.
- For Western blot, the predicted molecular weight is ~66 kDa; consider using reducing conditions and appropriate positive controls (e.g., human, mouse, or rat tissue lysates).
- For immunohistochemistry (IHC-P), validate in tissue sections from colon, breast, or lung tumors, where MMP14 is known to be expressed.
- For flow cytometry, note the cell surface localization; fixation and permeabilization protocols may influence detection sensitivity.
- ELISA applications may be suitable for detecting MMP14 in biological samples; consider sandwich pair optimization.
