Biological Background: MMP14 Function and Localization
- MMP14 (Matrix metalloproteinase-14) is also known as MT1-MMP, MT-MMP 1, and MMP-X1, a transmembrane zinc-dependent endopeptidase.
- It functions as a key pericellular collagenase, degrading various extracellular matrix components such as collagen, which is essential for connective tissue remodeling during development. Related references: PMID:8015608

Experimental Guidance and Technical Tips
- The immunogen spans amino acids 21-538, covering the entire extracellular domain including the catalytic and hemopexin domains, which may yield antibodies recognizing multiple epitopes and potentially the pro-form and active enzyme.
- For Western blotting, consider using reducing and denaturing conditions to resolve the predicted 66kDa protein; note that glycosylation or proteolytic processing may produce additional bands.
- In immunofluorescence (IF/ICC) and IF-P, membrane staining is expected; permeabilization may be required for cytoplasmic detection.
- ELISA can be designed with this polyclonal antibody as capture or detection reagent, but appropriate validation in the sample matrix is essential.
