Biological Background: MMP13 Function and Localization
- Collagenase 3 (MMP-13) is a secreted matrix metalloproteinase encoded by the MMP13 gene, with a predicted molecular mass of 53.8 kDa and an observed ~54 kDa band in western blotting.
- It principally cleaves triple-helical fibrillar collagens, showing highest activity against soluble type II collagen, but also degrades types I, III, IV, X, and XIV, as well as fibronectin and tenascin.
- MMP13 belongs to the zinc- and calcium-dependent metalloprotease family; it requires calcium for stability and contains conserved domains typical of MMPs.
- The enzyme is secreted into the extracellular space and matrix, where it performs its catalytic functions.
- Expression is detected in fetal cartilage, hypertrophic chondrocytes, osteoblasts, periosteal cells, and activated T lymphocytes; it is also found in breast carcinoma tissue.
- Post-translational modifications include phosphorylation and glycosylation; the zymogen form contains a signal peptide and conserved disulfide bonds.
- MMP13 is essential for normal embryonic bone development, skeleton ossification, and bone fracture healing; it also promotes keratinocyte migration in wound healing and may facilitate tumor cell invasion.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide corresponding to residues 350–450 of human MMP13, which lies in a region likely conserved across the active enzyme.
- For western blotting, a single band near 54 kDa is typically observed under reducing conditions; validate expression in appropriate cell lines or tissue lysates.
- In immunofluorescence/ICC, signal is expected in the extracellular matrix and secretory pathway; consider co-staining with organelle markers.
- ELISA applications may require pairing with a suitable capture antibody; test linearity using recombinant MMP13 standards.
- Since MMP13 is a secreted protein, cell culture supernatants or tissue extracts are suitable sample types.
