- MMP13, also known as collagenase 3, is a member of the matrix metalloproteinase (MMP) family of zinc-dependent endopeptidases.
- It plays a crucial role in the degradation of extracellular matrix proteins, particularly fibrillar collagens such as type I, type II, and type III collagen.
- MMP13 cleaves triple helical collagens with highest activity towards soluble type II collagen, and also degrades collagen types IV, X, and XIV, fibronectin, and ACAN.
- Beyond matrix degradation, MMP13 can proteolytically activate growth factors like TGFB1 and degrade CCN2, influencing wound healing and tissue remodeling.
- Subcellularly, MMP13 is secreted into the extracellular space and matrix, where it functions in cartilage degradation, bone development, mineralization, and endochondral ossification.
- It is essential for normal embryonic bone development and bone fracture healing, and also contributes to keratinocyte migration during wound repair.
