- MMP10 (Stromelysin-2) is also known as matrix metalloproteinase-10 (MMP-10) or Transin-2, encoded by the MMP10 gene.
- It is a secreted zinc-dependent endopeptidase that degrades various extracellular matrix components, including fibronectin, gelatins of type I, III, IV, and V, and weakly collagens III, IV, and V.
- MMP10 can activate procollagenase, thereby participating in the proteolytic cascade that remodels the extracellular matrix.
- As a member of the matrix metalloproteinase family, it shares structural features such as a signal peptide, propeptide domain, and catalytic domain with conserved zinc-binding and calcium-binding sites.
- Subcellularly, MMP10 is secreted into the extracellular space and localizes to the extracellular matrix, consistent with its role in matrix turnover.
- The enzyme is synthesized as an inactive zymogen and requires proteolytic cleavage for activation, a regulatory mechanism typical of MMPs.
- Post-translational modifications include disulfide bond formation, and the protein contains characteristic repeat structures and metal-binding motifs essential for its hydrolytic activity.
Experimental Guidance and Technical Tips
- The immunogen corresponds to a region within the mature protein (amino acids 237–476), which may recognize both the pro-form and active enzyme without preference. Validate in your model system if active-form-specific detection is needed.
- For Western blotting, expect a band near 54 kDa (precursor) or slightly lower for the mature form after activation; reducing conditions are recommended.
- In immunohistochemistry (IHC-P), antigen retrieval using heat-mediated citrate or EDTA buffer may enhance signal; tissue expression levels can be variable, so appropriate controls are essential.
- For immunofluorescence/immunocytochemistry (IF/ICC), fixation and permeabilization protocols should be optimized, as MMP10 is secreted and may require a Golgi-blocking agent to enhance intracellular retention.
- When used in ELISA, the polyclonal nature can provide sensitive capture/detection, but cross-reactivity with other MMPs should be assessed given sequence homology within the family.
- As a polyclonal antibody, lot-to-lot consistency may vary; it is advisable to titrate each new lot and confirm reactivity across intended species (human, mouse, rat).
