Biological Background: MMACHC Function and Localization

  • MMACHC (cblC) is a cytosolic cobalamin chaperone and enzyme with dual activities: cyanocobalamin reductase (cyanide elimination) and alkylcobalamin dealkylase (glutathione transferase).
  • It catalyzes the reductive decyanation of cyanocobalamin (inactive dietary B12) to cob(II)alamin using FAD/FMN and NADPH, a critical step for producing active cofactors methylcobalamin and adenosylcobalamin. Related references: PMID:18779575, PMID:19700356, PMID:21697092
