Biological Background: MLL5/KMT2E Function and Localization

MLL5, also known as histone reader KMT2E, is an inactive histone-lysine N-methyltransferase 2E that acts as a chromatin-associated regulator. It binds tri-methylated histone H3 at Lys-4 (H3K4me3) via its PHD finger domain, mediating chromatin interaction downstream of transcriptional start sites of active genes. Subcellularly, MLL5 localizes to the nucleus (nucleoplasm and nuclear speckles), but can also be found in the cytoplasm, centrosome, and cell membrane depending on the isoform and cellular context.

The protein is widely expressed in both adult and fetal tissues, with highest levels in fetal thymus and kidney, and adult hematopoietic tissues, jejunum, and cerebellum. Experimental Guidance and Technical Tips
- The immunogen covers amino acids 200-500, a region containing the PHD finger domain. This antibody may be particularly suitable for detecting chromatin-bound MLL5 in ChIP-related applications if adapted.
- For Western blot, the predicted molecular weight is ~205 kDa; consider using a low-percentage gel and extended transfer times. Validate specificity with appropriate controls, as multiple isoforms exist.
- In immunofluorescence and immunohistochemistry, MLL5 can display nuclear speckle or centrosomal staining patterns depending on cell type and fixation. Include permeabilization steps and co-staining with organelle markers.
- For ELISA, use the recombinant protein immunogen as a positive coating antigen to establish assay conditions. Cross-reactivity with mouse and rat MLL5 may allow multi-species testing.
