Biological Background: MLL5/KMT2E Function and Localization

  • MLL5, also known as histone reader KMT2E, is an inactive histone-lysine N-methyltransferase 2E that acts as a chromatin-associated regulator. It binds tri-methylated histone H3 at Lys-4 (H3K4me3) via its PHD finger domain, mediating chromatin interaction downstream of transcriptional start sites of active genes.   • It plays a key role in hematopoiesis, regulating terminal myeloid differentiation and hematopoietic stem cell self-renewal through a mechanism involving DNA methylation (By similarity).

  • MLL5 participates in cell cycle regulation at multiple stages, including G1/S transition, S phase progression, and mitotic entry. It is recruited to E2F1-responsive promoters by HCFC1, where it stimulates H3K4me3 and transcriptional activation, facilitating G1 to S phase transition.   • For Western blot, the predicted molecular weight is ~205 kDa; consider using a low-percentage gel and extended transfer times. Validate specificity with appropriate controls, as multiple isoforms exist.
  • In immunofluorescence and immunohistochemistry, MLL5 can display nuclear speckle or centrosomal staining patterns depending on cell type and fixation. Include permeabilization steps and co-staining with organelle markers.
  • For ELISA, use the recombinant protein immunogen as a positive coating antigen to establish assay conditions. Cross-reactivity with mouse and rat MLL5 may allow multi-species testing.
