Biological Background: MLF1 Function and Localization
- MLF1 (Myeloid leukemia factor 1), also known as myelodysplasia-myeloid leukemia factor 1, is encoded by the MLF1 gene (GeneID: 4291).
- Plays a critical role in lineage commitment of primary hemopoietic progenitors: restricts erythroid formation and promotes myeloid formation.
- Interferes with erythropoietin-induced erythroid terminal differentiation by preventing cell cycle exit through suppression of CDKN1B/p27Kip1 levels.
- Suppresses COP1 activity via CSN3, thereby activating p53 and inducing cell cycle arrest.
- Binds DNA and may function as a transcription factor, affecting expression of multiple downstream genes.
- Localizes to the cytoplasm and nucleus, and is also found at the cilium and basal body.
- Tissue expression is highest in testis, ovary, skeletal muscle, heart, kidney, and colon; lower in spleen, thymus, and peripheral blood leukocytes.
- Post-translational modifications include phosphorylation; also undergoes alternative splicing and chromosomal rearrangement in some leukemias.

- The immunogen is a recombinant fusion protein covering amino acids 1-268 of human MLF1. For Western blot, the expected band is approximately 31 kDa; consider using a positive control lysate from human or mouse cell lines.
- For IF/ICC, fixation and permeabilization conditions may need optimization to detect MLF1 in its various localizations (cytoplasm, nucleus, cilia). Co-staining with compartment-specific markers is recommended.
- For ELISA, the antibody can be used with the immunogen as a capture or detection reagent; pairing with a matched detection antibody may enhance sensitivity.
