Biological Background: MKL1 Function and Localization
- MKL1 (Myocardin-related transcription factor A), also known as MRTF-A, MAL, BSAC, is a member of the myocardin family of transcriptional coactivators.
- It associates with serum response factor (SRF) to drive expression of cytoskeletal genes, playing key roles in development, morphogenesis, and cell migration. Related references: PMID:26224645
- MKL1 activity is tightly regulated by the Rho GTPase signaling pathway through changes in cellular G-actin and F-actin levels; it binds G-actin via its RPEL repeats, and nuclear F-actin also modulates activity.
- Predominantly localizes to the cytoplasm, but translocates to the nucleus upon actin polymerization changes, enabling SRF-mediated transcription.

- In immunofluorescence (IF/ICC), monitor MKL1 nuclear translocation as a readout for SRF pathway activation; serum stimulation or Rho activation can be used as positive controls.
