Biological Background: MKL1 Function and Localization

  • MKL1 (Myocardin-related transcription factor A, also known as MRTF-A, MAL, or BSAC) is encoded by the MRTFA gene and belongs to the myocardin family of transcriptional coactivators.
  • It associates with serum response factor (SRF) to regulate expression of genes controlling cytoskeletal dynamics during development, morphogenesis, and cell migration. Related references: PMID:26224645
  • Activity is modulated by Rho GTPase signaling and cellular actin dynamics; MKL1 binds G-actin via its RPEL repeats, and nuclear F-actin levels further regulate the MRTFA-SRF complex.
  • MKL1 localizes to both the cytoplasm and nucleus, undergoing nucleocytoplasmic shuttling in response to stimuli.

  • For Western blot (WB), the expected band is around 99 kDa; consider using whole-cell lysates from human, mouse, or rat cell lines, and include appropriate loading controls.
  • For ELISA applications, this antibody may be used as a capture or detection reagent; pairing with a specific MKL1 monoclonal antibody should be evaluated in the relevant sample system.
  • Cross-reactivity with mouse and rat has been confirmed by supplier, but antibody performance in other applications (IHC, IF, etc.) has not been validated; users should perform their own optimization.
