Biological Background: MITF Function and Localization
- Transcription factor that acts as a master regulator of melanocyte survival and differentiation as well as melanosome biogenesis.
Related references: PMID:10587587, PMID:22647378, PMID:27889061 - Binds to M-boxes (5'-TCATGTG-3') and symmetrical DNA sequences (E-boxes) (5'-CACGTG-3') found in the promoter of pigmentation genes, such as tyrosinase (TYR).
Related references: PMID:10587587, PMID:22647378, PMID:27889061 - Involved in the cellular response to amino acid availability by acting downstream of MTOR: in the presence of nutrients, MITF phosphorylation by MTOR promotes its inactivation; upon starvation or lysosomal stress, inhibition of MTOR induces MITF dephosphorylation, resulting in transcription factor activity.
Related references: PMID:36608670 - Plays an important role in melanocyte development by regulating the expression of tyrosinase (TYR) and tyrosinase-related protein 1 (TYRP1).
Related references: PMID:10587587, PMID:22647378, PMID:27889061 - Critical in the differentiation of various cell types, such as neural crest-derived melanocytes, mast cells, osteoclasts, and optic cup-derived retinal pigment epithelium.
Related references: PMID:10587587, PMID:22647378, PMID:27889061 - Localizes to the nucleus, cytoplasm, and lysosome membrane, reflecting its dynamic regulatory roles.
- Expressed in melanocytes (at protein level), retinal pigment epithelium, brain, placenta, and kidney.
Related references: PMID:9647758, PMID:10578055 - MITF mutations are associated with Waardenburg syndrome type 2A, Tietz syndrome (albinism-deafness), and microphthalmia, highlighting its developmental importance.
Experimental Guidance and Technical Tips
- The immunogen (aa 1-280) targets the N-terminal region of human MITF, which may recognize multiple isoforms if this region is conserved.
- Predicted molecular weight is ~59 kDa; however, UniProt reports a calculated mass of ~58.8 kDa. Post-translational modifications may cause band shifts in WB.
- For Western blot, standard protocols for polyclonal antibodies can be applied. Validate the antibody in relevant cell or tissue lysates expressing MITF (e.g., melanocyte lines, retinal pigment epithelium).
- For ELISA, consider using recombinant MITF protein or peptide as capture antigen; titer and specificity should be determined experimentally.
- Cross-reactivity with mouse indicates potential use in mouse models, but validation in mouse tissue extracts is recommended.
