Biological Background: MITF Function and Localization

  • MITF (Microphthalmia-associated transcription factor) is a Class E basic helix-loop-helix protein (bHLHe32) that functions as a master regulator of melanocyte survival, differentiation, and melanosome biogenesis. Related references: PMID:10587587, PMID:22647378, PMID:27889061
  • It binds to M-box (5'-TCATGTG-3') and E-box (5'-CACGTG-3') DNA elements in the promoters of pigmentation genes, including tyrosinase (TYR) and tyrosinase-related protein 1 (TYRP1), thereby regulating melanin synthesis. Related references: PMID:10587587, PMID:22647378, PMID:27889061
  • MITF activity is coupled to nutrient sensing: in the presence of amino acids, MTOR-mediated phosphorylation inactivates MITF, whereas starvation or lysosomal stress leads to its dephosphorylation and activation, linking metabolism to transcription. Related references: PMID:36608670
  • The protein localizes mainly to the nucleus but can also be found in the cytoplasm and on the lysosome membrane, indicative of multiple regulatory compartments.
  • Expression is highest in melanocytes and retinal pigment epithelium, with additional presence in brain, placenta, and kidney. Related references: PMID:9647758, PMID:10578055
  • Multiple isoforms are generated by alternative splicing, and mutations in the MITF gene are linked to Waardenburg syndrome type 2A, Tietz albinism-deafness syndrome, and COMMAD syndrome, underscoring its developmental importance.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to the C-terminal region (aa 402–526) of human MITF, which is conserved and may contribute to the antibody's cross-reactivity with rat MITF.
  • For Western blot, an expected band around 59 kDa is predicted based on the canonical isoform; however, alternative splicing may produce additional bands.
  • In immunofluorescence/immunocytochemistry, nuclear and potentially cytoplasmic staining should be anticipated given MITF's subcellular localization.
  • This rabbit monoclonal format provides high batch-to-batch consistency, making it suitable for reproducible ELISA-based detection methods.
  • Perform appropriate controls (e.g., siRNA knockdown or overexpression) to confirm specificity in the relevant tissue or cell type.
