- Mineralocorticoid receptor (MR, NR3C2) is a nuclear receptor that mediates the effects of mineralocorticoids like aldosterone and glucocorticoids such as cortisol. It binds to mineralocorticoid response elements (MREs) in DNA to regulate gene transcription.
- Activation of MR increases ion and water transport in epithelial tissues, thereby raising extracellular fluid volume and blood pressure while lowering serum potassium levels.
- MR belongs to the nuclear receptor subfamily 3 group C, featuring a zinc-finger DNA-binding domain and a C-terminal steroid-binding domain.
- The receptor is expressed ubiquitously, with highest levels in kidney distal tubules, cortical collecting ducts, and sweat glands; lower expression is found in cardiomyocytes, epidermis, and colon enterocytes.
- Subcellularly, MR shuttles between the cytoplasm and nucleus, and is also associated with the endoplasmic reticulum membrane upon ligand activation.
- Post-translational modifications include phosphorylation, and alternative splicing generates isoforms with potentially distinct functional properties.
