Biological Background: MIF Function and Localization
- MIF, also designated as glycosylation-inhibiting factor (GIF), L-dopachrome isomerase, and phenylpyruvate tautomerase, is encoded by the MIF gene (GLIF, MMIF).
- As a pro-inflammatory cytokine, MIF participates in innate immunity against bacterial pathogens and modulates macrophage activity during host defense. Related references: PMID:15908412 PMID:17443469 PMID:23776208
- MIF counteracts the anti-inflammatory effects of glucocorticoids. Related references: PMID:15908412 PMID:17443469 PMID:23776208
- The protein exhibits in vitro tautomerase activity towards phenylpyruvate and dopachrome, although the physiological significance of these activities is not firmly established. Related references: PMID:11439086 PMID:17526494
- Subcellularly, MIF is localized to both the cytoplasm and the secretory pathway.
- Key post-translational modifications include acetylation, and the protein is a member of the reference proteome.
Experimental Guidance and Technical Tips
- The immunogen corresponds to full-length human MIF (aa 1-115); the polyclonal antibody may recognize both native and denatured protein conformations.
- For Western blotting, consider using whole cell lysates from human, mouse, or rat tissues/cells, given the reported cross-reactivity.
- For immunofluorescence/ICC, validate the antibody in relevant cell lines under appropriate fixation and permeabilization conditions.
- ELISA applications could be explored for quantitative detection of MIF; note that MIF is a secreted protein, so culture supernatants may be a suitable sample type.
- As with any polyclonal antibody, lot-to-lot consistency should be verified; consider including appropriate controls in each experiment.
