Biological Background: MGAT3 Function and Localization

  • MGAT3 encodes beta-1,4-mannosyl-glycoprotein 4-beta-N-acetylglucosaminyltransferase (also known as GNT-III or N-acetylglucosaminyltransferase III), a key glycosyltransferase that adds a bisecting N-acetylglucosamine (GlcNAc) to the beta-linked mannose of the trimannosyl core of N-linked glycans, thereby modifying glycoprotein oligosaccharide structure and function.
  • The addition of this bisecting GlcNAc residue suppresses further processing and elongation of N-glycans, precluding the formation of beta-1,6 GlcNAc branching catalyzed by MGAT5, as MGAT5 cannot use the bisected oligosaccharide as a substrate (PubMed:19403558). Related references: PMID:19403558
  • MGAT3-catalyzed bisecting GlcNAc modifications impact cell adhesion and migration: addition to CDH1/E-cadherin modulates its cell membrane localization (PubMed:19403558), and inhibition of NeuAc-alpha-2,3-Gal-beta-1,4-GlcNAc- formation by MGAT3 regulates sialylation and cell migration (PubMed:26801611). Related references: PMID:19403558 PMID:26801611
  • In the brain, MGAT3 adds bisecting GlcNAc to BACE1, blocking its lysosomal targeting under oxidative stress and increasing its localization to early endosomes, which enhances APP cleavage and amyloid beta generation.
  • MGAT3 modifies IgG Fc N-glycans by adding bisecting GlcNAc to complex-type N-linked glycans, affecting antibody-dependent cellular cytotoxicity (ADCC) and potentially therapeutic antibody functions.
  • The enzyme is localized to the Golgi apparatus membrane, specifically the Golgi stack membrane, consistent with its role in late-stage N-glycan processing.
  • Alternative names include GNT3, GNT-III, and N-acetylglucosaminyltransferase III; the protein is predicted to contain a transmembrane helix and a signal-anchor.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to amino acids 364–533, a region accessible for antibody generation; consider using this antibody in Western blot under reducing conditions, where a major band around 61kDa may be observed.
  • For ELISA, optimize coating conditions and validate in the intended sample matrix, as glycosylation patterns and target abundance may vary between tissues or cell lines.
  • Given the reported cross-reactivity with mouse and rat, validate target specificity in these species by sequence alignment or additional controls, especially if used in non-human samples.
  • Since MGAT3 is a Golgi-resident protein, consider appropriate sample lysis and fractionation protocols to enrich membrane compartments for more robust detection.
