Biological Background: MGAT3 Function and Localization
- MGAT3, also known as GNT-III (N-acetylglucosaminyltransferase III), is a glycosyltransferase that catalyzes the addition of N-acetylglucosamine (GlcNAc) in beta-1,4 linkage to the beta-linked mannose of the trimannosyl core of N-linked sugar chains, forming bisecting GlcNAc.
- The bisecting GlcNAc moiety alters N-glycan conformation and suppresses further processing, including the formation of beta-1,6 GlcNAc branching by MGAT5, which cannot utilize bisected substrates. Related references: PMID:19403558
- MGAT3-mediated bisecting GlcNAc addition to CDH1/E-cadherin modulates its cell membrane localization. Related references: PMID:19403558
- It inhibits the formation of NeuAc-alpha-2,3-Gal-beta-1,4-GlcNAc- structures, thereby regulating sialylation levels and influencing cell migration. Related references: PMID:26801611
- In the brain, MGAT3 adds bisecting GlcNAc to BACE1, which blocks its lysosomal targeting under oxidative stress, increasing early endosome localization and APP cleavage (By similarity).
- MGAT3 modifies complex-type N-linked glycans on IgG Fc regions, converting various core glycoforms; galactosylation on the alpha(1->6) Man branch is permissive.
- Located in the Golgi stack membrane, MGAT3 is a single-pass type II membrane protein with a signal-anchor topology.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide corresponding to amino acids 381-480 of human MGAT3. Verify antibody recognition of the full-length endogenous protein under your experimental conditions, as peptide antibodies may not always detect native conformations.
- For Western blotting, consider using reducing conditions and a molecular weight marker around 61 kDa. Validate in the relevant sample system, as expression levels and post-translational modifications can vary.
- In ELISA applications, optimize coating antigen and antibody concentrations to achieve the desired sensitivity.
