Biological Background: MFSD2A/NLS1 Function and Localization
- Encodes a sodium-dependent lysophosphatidylcholine (LPC) symporter that is essential for blood-brain barrier (BBB) formation and function, mediating LPC transport across the endothelium. Related references: PMID:24828040, PMID:32572202, PMID:34135507
- Serves as the major route for brain uptake of docosahexaenoic acid (DHA), an omega-3 fatty acid critical for neurodevelopment, by transporting LPC-DHA rather than unesterified DHA. Related references: PMID:26005868, PMID:34135507
- Transports LPC species with long-chain fatty acids (minimum 14-carbon acyl chain), such as LPC-oleate and LPC-palmitate, but does not transport free fatty acids.
- In placenta, acts as a receptor for the endogenous retroviral envelope protein syncytin-2 (ERVFRD-1), a function required for trophoblast cell fusion during syncytiotrophoblast formation. Related references: PMID:18988732, PMID:23177091

Experimental Guidance and Technical Tips
- The immunogen is a recombinant human protein; therefore, consider using recombinant MFSD2A or peptide fragments as positive controls in ELISA and WB.
- For Western blot, prepare whole-cell or membrane-enriched lysates from human, mouse, or rat brain microvessel endothelial cells or placental tissue where MFSD2A is expressed.
- Due to the predicted molecular weight of approximately 60 kDa, include appropriate molecular weight markers and optimize transfer conditions for proteins of this size.
- As with any antibody, titrate the working concentration for your specific sample type and application; cross-reactivity with rat and mouse has been reported but validate in your system.
- Note that MFSD2A may undergo glycosylation and other modifications; observed band sizes may differ slightly from the theoretical 60 kDa.
