Biological Background: Mfsd2a Function and Localization

  • Mfsd2a (Major facilitator superfamily domain-containing protein 2A), also known as NLS1, is a sodium-dependent lysophosphatidylcholine (LPC) symporter essential for blood-brain barrier formation and function. Related references: PMID:18694395, PMID:23209793, PMID:24828040

  • It specifically transports LPC into the brain, which is the major route for docosahexaenoic acid (DHA) entry, an omega-3 fatty acid critical for normal brain growth and cognitive function. Related references: PMID:24828040, PMID:24828044, PMID:34349262

  • Mfsd2a transports LPC species with acyl chain lengths of at least 14 carbons, such as LPC oleate and LPC palmitate, but does not transport unesterified DHA. Related references: PMID:24828044, PMID:34349262

  • It belongs to the major facilitator superfamily (MFS) and is predicted to contain 12 transmembrane helices, consistent with its transporter function.

  • Subcellularly, Mfsd2a localizes to the cell membrane and endoplasmic reticulum membrane, supporting its role in lipid uptake and intracellular trafficking.

  • The protein is widely expressed but enriched in brain micro-vessel endothelium forming the blood-brain barrier, and exhibits circadian oscillatory expression in brown adipose tissue and liver.

  • Mfsd2a is not required for central nervous system vascular morphogenesis, highlighting its specific transport rather than developmental role. Related references: PMID:24828040

Experimental Guidance and Technical Tips

  • For Western blot (WB), use protein lysates from brain, liver, or brown adipose tissue, where Mfsd2a is highly expressed. Consider enrichment for membrane fractions to enhance detection.
  • Validate antibody specificity by siRNA knockdown or CRISPR-mediated knockout of Mfsd2a in relevant cell lines.
  • For ELISA, optimize the coating antigen using recombinant Mfsd2a protein or a peptide containing the immunogen region (amino acids 170-245).
  • Due to the polyclonal nature, assess batch-to-batch variability for reproducible quantitative assays.
  • Ensure cold shipping and storage at recommended temperatures to maintain antibody activity.
