Biological Background: MFSD2A Function and Localization
- MFSD2A is a sodium-dependent lysophosphatidylcholine (LPC) symporter essential for blood-brain barrier (BBB) formation and function. Related references: PMID:24828040 PMID:32572202 PMID:34135507
- It transports LPC into the brain, which constitutes the major pathway for docosahexaenoic acid (DHA) delivery critical for normal brain growth and cognitive function. Related references: PMID:26005868 PMID:34135507
- The protein belongs to the major facilitator superfamily (MFS) and is specifically expressed in endothelial cells of the BBB.
- Subcellularly, MFSD2A localizes to the cell membrane and endoplasmic reticulum membrane.

- For Western blot, consider using whole cell lysates from human, mouse, or rat tissues known to express MFSD2A (e.g., brain, placenta). The predicted molecular weight is approximately 60 kDa; however, glycosylation may cause a shift in migration.
- For ELISA, titrate the antibody against the immunogen or native protein to determine the optimal working concentration. Validate specificity with appropriate controls.
- Always include a positive control (e.g., human brain microvascular endothelial cells or placental tissue) and a negative control to confirm detection.
