Biological Background: Lactadherin (MFGE8) Function and Localization
- Lactadherin (MFGE8) is a secreted and membrane-associated glycoprotein also known as breast epithelial antigen BA46, HMFG, MFGM, SED1, and EDIL1.
- It plays a critical role in maintaining intestinal epithelial homeostasis and promoting mucosal healing, partly through VEGF-dependent neovascularization.
- MFGE8 acts as a specific ligand for integrin receptors alpha-v/beta-3 and alpha-v/beta-5, and also binds phosphatidylserine on apoptotic cells, facilitating their phagocytic clearance.
- The protein is involved in gamete interaction as a zona pellucida-binding protein, suggesting a role in fertilization.
- It is a main constituent of aortic medial amyloid.
- MFGE8 is expressed on mammary epithelial cell surfaces and in the aortic media; it is overexpressed in several carcinomas.
- The protein undergoes alternative splicing, glycosylation, and phosphorylation; contains EGF-like domains and disulfide bonds.
- Subcellularly, it localizes to the membrane, is secreted, and found in cytoplasmic vesicles, including secretory vesicles and acrosome membranes.

- The immunogen covers amino acids 1-200 of human MFGE8 (NP_005919.2), which includes the N-terminal region; this sequence may contain the signal peptide and part of the mature protein. Consider the potential impact on antibody recognition of secreted versus membrane-bound forms.
- For Western blot, the predicted molecular weight is ~43 kDa; however, due to glycosylation and alternative splicing, additional bands may be observed. Validate using relevant cell lysates or recombinant protein controls.
- In immunofluorescence (IF/ICC) and immunohistochemistry (IHC-P), note that MFGE8 localizes to the cell membrane, secreted vesicles, and cytoplasmic compartments; include appropriate permeabilization and fixation protocols for detection.
- The antibody cross-reacts with human, mouse, and rat; thus, it is suitable for studies in these species, but validation in each species/model is recommended.
