Biological Background: Lactadherin (MFGE8) Function and Localization

• Lactadherin (MFGE8) is a secreted and membrane-associated glycoprotein also known as breast epithelial antigen BA46, HMFG, MFGM, SED1, and EDIL1.
• It plays a critical role in maintaining intestinal epithelial homeostasis and promoting mucosal healing, partly through VEGF-dependent neovascularization.
• MFGE8 acts as a specific ligand for integrin receptors alpha-v/beta-3 and alpha-v/beta-5, and also binds phosphatidylserine on apoptotic cells, facilitating their phagocytic clearance.
• The protein is involved in gamete interaction as a zona pellucida-binding protein, suggesting a role in fertilization.
• It is a main constituent of aortic medial amyloid.

• For Western blot, the predicted molecular weight is ~43 kDa; however, due to glycosylation and alternative splicing, additional bands may be observed. Validate using relevant cell lysates or recombinant protein controls.
• In immunofluorescence (IF/ICC) and immunohistochemistry (IHC-P), note that MFGE8 localizes to the cell membrane, secreted vesicles, and cytoplasmic compartments; include appropriate permeabilization and fixation protocols for detection.
