Biological Background: METTL3 Function and Localization

  • METTL3 (Methyltransferase-like protein 3) is the catalytic subunit of the N6-adenosine-methyltransferase complex, which includes METTL14 as a regulatory partner. Related references: PMID:27281194 PMID:27373337 PMID:27627798
  • The heterodimer deposits N6-methyladenosine (m6A) on RNA at the consensus sequence 5'-[AG]GAC-3', thereby regulating mRNA stability, processing, translation efficiency, and editing. Related references: PMID:22575960 PMID:24284625 PMID:25719671
  • m6A modification acts as a key regulator of mRNA stability; methylation promotes mRNA destabilization and degradation, influencing processes such as embryonic stem cell differentiation and the circadian clock. m6A also regulates circadian hepatic lipid metabolism. Related references: PMID:30428350
  • In response to UV-induced DNA damage, METTL3 rapidly methylates poly(A) transcripts at damage sites, facilitating the recruitment of DNA repair polymerase POLK. Related references: PMID:28297716
  • METTL3-mediated m6A methylation plays critical roles in immunity: it inhibits the type I interferon response by methylating IFNB, and dampens innate immune activation during SARS-CoV-2 infection by modifying viral RNA. Related references: PMID:30559377 PMID:33961823
  • The enzyme also methylates primary miRNAs (pri-miRNAs), marking them for recognition and processing by DGCR8, thus linking m6A modification to miRNA biogenesis. Related references: PMID:25799998
  • Beyond its catalytic activity, METTL3 can promote mRNA translation by interacting with translation initiation factors in the cytoplasm, a function implicated in cancer cell proliferation. Related references: PMID:27117702
  • METTL3 localizes to the nucleus, nuclear speckles, and cytoplasm, and is widely expressed at low levels, with higher expression in spleen, thymus, prostate, testis, ovary, small intestine, colon, and peripheral blood leukocytes.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to the N-terminal region (amino acids 1–156) of human METTL3, targeting a conserved domain; this may be suitable for detecting METTL3 across various applications and species.
  • For Western blot (WB), a band at approximately 64 kDa is expected. Validation of expression levels in your specific sample system is recommended.
  • In immunohistochemistry (IHC-P), consider antigen retrieval optimization and inclusion of appropriate tissue controls known to express METTL3 (e.g., testis, spleen).
  • For immunoprecipitation (IP), verify compatibility with the native METTL3-METTL14 complex and confirm capture efficiency under your experimental conditions.
  • Cross-reactivity with mouse and rat is indicated; confirm target sequence homology in your model organism before use.
