Biological Background: Mesothelin Function and Localization

  • Mesothelin is a cell surface glycoprotein encoded by the MSLN gene, also known as MPF (megakaryocyte-potentiating factor) or CAK1 antigen.
  • The full-length protein is synthesized as a 71 kDa precursor that undergoes proteolytic cleavage to yield a 31 kDa N-terminal secreted form (megakaryocyte-potentiating factor, MPF) and a 40 kDa C-terminal membrane-bound mesothelin.
  • Membrane-anchored mesothelin is attached to the cell surface via a glycosylphosphatidylinositol (GPI) anchor and is implicated in cell adhesion.
  • MPF is secreted and has been shown to potentiate megakaryocyte colony formation in vitro, suggesting a role in hematopoiesis.
  • Mesothelin is predominantly expressed in mesothelial cells lining the pleura, pericardium, and peritoneum, and is also found at lower levels in lung, heart, placenta, and kidney.
  • It is highly overexpressed in several human cancers, including mesotheliomas, ovarian cancers, and some squamous cell carcinomas, making it a target for cancer diagnostics and therapeutics.
  • The protein undergoes several post-translational modifications, including glycosylation, disulfide bond formation, and phosphorylation, which are critical for its stability and function.
  • Subcellularly, mesothelin localizes to the cell membrane, the Golgi apparatus, and is also found in secreted forms.

Experimental Guidance and Technical Tips

  • The immunogen encompasses the C-terminal region (amino acids 296–606) of human mesothelin, which corresponds to the mature membrane-bound form; therefore, the antibody is expected to detect both the full-length precursor and the cleaved membrane-bound subunit in Western blotting.
  • For Western blot, consider using reducing conditions, as disulfide bonds may affect migration; the predicted molecular weight of the mature membrane mesothelin is approximately 40 kDa, but bands at higher molecular weights may be observed due to glycosylation.
  • In immunohistochemistry (IHC-P), optimize antigen retrieval and validate in mesothelial or tumor tissue sections known to overexpress mesothelin, such as mesothelioma or ovarian cancer biopsies.
  • For ELISA, pair with a capture antibody targeting a non-overlapping epitope to assess mesothelin levels in serum or cell culture supernatants; note that the soluble MPF form may also be detected if the epitope is present in the N-terminal region.
