Biological Background: Tyrosine-protein kinase Mer (MERTK) Function and Localization

  • MERTK is a receptor tyrosine kinase that transduces signals from the extracellular matrix by binding ligands like LGALS3, TUB, TULP1, and GAS6, regulating cell survival, migration, differentiation, and phagocytosis of apoptotic cells (efferocytosis).
  • Ligand binding induces autophosphorylation, creating docking sites for downstream signaling molecules such as GRB2 or PLCG2, leading to phosphorylation of MAPK1, MAPK2, FAK/PTK2, or RAC1.
  • MERTK plays a crucial role in macrophage clearance of apoptotic cells, platelet aggregation, cytoskeleton reorganization, and engulfment. Related references: PMID:32640697
  • In the retinal pigment epithelium (RPE), MERTK functions as a regulator of rod outer segment fragments phagocytosis, essential for photoreceptor maintenance.
  • MERTK inhibits Toll-like receptor (TLR)-mediated innate immune responses by activating STAT1, which induces suppressors of cytokine signaling SOCS1 and SOCS3.
  • The protein is localized to the cell membrane.
  • Tissue specificity: Not expressed in normal B- and T-lymphocytes but expressed in neoplastic B- and T-cell lines; highly expressed in testis, ovary, prostate, lung, kidney, with lower expression in spleen, small intestine, colon, liver.
  • Additional features: contains immunoglobulin domains, is glycosylated, phosphorylated, and has disulfide bonds; it is a proto-oncogene associated with disease variants in retinitis pigmentosa.

Experimental Guidance and Technical Tips

  • The antibody was raised against the N-terminal region (amino acids 1-100) of human MERTK, a region that may be well-exposed in denaturing Western blot conditions; consider optimizing the primary antibody dilution and incubation conditions for your specific sample type.
  • For Western blot (WB), a protein band around 110 kDa is expected; validate the signal in MERTK-expressing cell lines or tissues, keeping in mind its reported expression pattern.
  • For ELISA, plate coating with recombinant MERTK or cell lysates may be used; cross-reactivity is declared against human MERTK, and reactivity with other species should be independently verified if required.
  • The antibody is supplied in liquid form with ice bag shipping; store at -20°C or as recommended to maintain stability.
