Biological Background: MEPE Function and Localization
- MEPE (Matrix extracellular phosphoglycoprotein), also known as OF45 or osteoregulin, is a secreted extracellular matrix phosphoglycoprotein involved in biomineralization.
- Promotes renal phosphate excretion and inhibits intestinal phosphate absorption. Related references: PMID:14962809 PMID:19005008
- Promotes bone mineralization by osteoblasts and cartilage mineralization by chondrocytes. Related references: PMID:18162525 PMID:19998030 PMID:22766095
- Regulates the mineralization of the extracellular matrix of the craniofacial complex, including teeth, bone, and cartilage (By similarity).
- Promotes dental pulp stem cell proliferation and differentiation. Related references: PMID:22341070
- Detected in urine and expressed by osteoblasts, dental pulp stem cells, mesenchymal cells in dental papilla and pulp, and odontoblasts.
- Localizes to the extracellular space and extracellular matrix; secreted protein.
- Subject to post-translational modifications: glycoprotein, phosphoprotein, proteoglycan.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 90-256 of human MEPE, a region likely to produce antibodies recognizing multiple epitopes due to the polyclonal nature.
- For Western blot (WB), the expected molecular weight is approximately 58 kDa. Validate using appropriate positive controls such as osteoblast or dental pulp cell lysates.
- For ELISA, consider using the recombinant MEPE protein as a standard and coating antigen; optimal working concentrations should be determined experimentally.
- Cross-reactivity is indicated for human, mouse, and rat; verify in your specific sample matrix if species other than those listed are used.
